Trial

ACCELERATE

Eli Lilly (evacetrapib) · NCT01687998 · 1 October 2012

Status Terminated for futility, 2015

12,092 patients with high-risk vascular disease randomised to evacetrapib or placebo, stopped early for futility. HDL-C more than doubled, LDL-C fell, and the event curves stayed together. The trial that ended the simple HDL hypothesis.

ACCELERATE is the cleanest experiment in the field: a drug that produced exactly the lipid changes the hypothesis called for, in a large high-risk population, with no effect on outcomes and no safety signal to blame. After it, arguing for CETP inhibition on HDL-C grounds became untenable.
