Trial

dal-OUTCOMES

Hoffmann-La Roche (dalcetrapib) · NCT00658515 · 1 April 2008

Status Completed 2012; neutral

15,871 patients randomised after a recent acute coronary syndrome to dalcetrapib or placebo. HDL-C rose by about a third, apoB barely moved, and there was no effect on cardiovascular outcomes. The first large trial to show that raising HDL-C with a CETP agent does not translate into benefit.

In hindsight dal-OUTCOMES was a test of the wrong mechanism: dalcetrapib modulates CETP rather than inhibiting it strongly, and leaves apoB largely intact. Its neutrality was read at the time as a verdict on the class; it is better read as a verdict on HDL-C as a target.
