6,147 patients selected for the ADCY9 genotype that a dal-OUTCOMES pharmacogenomic analysis suggested marked responders, randomised to dalcetrapib or placebo after an acute coronary syndrome. The first prospective test of a genotype-first hypothesis in this class.
dal-GenE exists because a retrospective genetic analysis of a neutral trial produced a striking interaction signal at the ADCY9 locus. Whether that signal is real is the question; a further genotype-defined trial (NCT05918861) is recruiting, which tells you the sponsor does not consider the matter closed.
